目的探讨HSD17B4基因在食管癌中的变化及其可能的意义。方法采用逆转录聚合酶链式反应(RT-PCR),分析HSD17B4基因mRNA表达状态,同时运用PCR银染技术,检测HSD17B4基因内微卫星标记D5S1384在食管癌组织基因组DNA水平的缺失。结果40例食管癌及其癌旁组织标本微卫星标记D5S1384分析发现,26例为信息个体,12例肿瘤组织存在D5S1384的杂合性丢失,丢失率为46.2%。HSD17B4基因mRNA表达下调的比例为62.5%(25/40)。结论HSD17B4可能是5q23区域内的候选食管癌抑制基因。
Objective To investigate the changes of HSD17B4 gene in esophageal cancer and its possible significance. Methods Reverse transcriptase polymerase chain reaction (RT-PCR) was used to analyze the expression of HSD17B4 gene mRNA. The PCR-silver staining technique was used to detect the deletion of genomic DNA in HSD17B4 microsatellite marker D5S1384. Results Microsatellite markers D5S1384 in 40 cases of esophageal cancer and its adjacent tissues showed that heterozygosity of D5S1384 was lost in 46 cases (26.2%) and loss of heterozygosity in D5S1384 (12 cases). The down-regulation of HSD17B4 mRNA was 62.5% (25/40). Conclusion HSD17B4 may be a candidate suppressor of esophageal cancer in the 5q23 region.
