目的 建立 3种新型α1 受体阻断剂的手性流动相HPLC分离与半制备方法。方法 通过探讨有机相、手性添加剂、流动相的pH、扫尾剂、反相固定相对手性分离的影响 ,选择最佳的手性分离条件。结果 基线分离了 3种新型α1 受体阻断剂对映体 ,并制备了毫克级样品。结论 所建立的分离与制备方法可用于该类新药的手性研究与开发
OBJECTIVE To establish a chiral HPLC separation and semi-preparative method for three novel α1 blockers. Methods The optimum conditions of chiral separation were selected by studying the effects of organic phase, chiral additives, pH of mobile phase, anti-tailing agent and reversed-phase stationary phase on chiral separation. Results Three enantiomers of the novel α1 receptor blockers were isolated at baseline and milligram samples were prepared. Conclusion The established separation and preparation methods can be used for the chiral research and development of these new drugs
