组织因子(tissue factor,TF)是生理性凝血过程与病理条件下血栓形成的启动因子,通过与凝血因子VII相互作用参与凝血过程,在血栓形成过程中起重要作用。归纳中药对血栓形成过程中组织因子的抑制作用,对于开发以TF为靶点的抗凝、抗血栓药物具有重要的意义。通过免疫组化、酶联免疫吸附法(ELISA)、反转录聚合酶链反应(RT-PCR)、发色底物法和免疫印迹法(Western blot)来检测单味中药和复方中药对血栓形成中TF蛋白、TF mRNA、蛋白激酶C(PKC)和核转录因子κB(NF-κB)表达的影响,结果表明单味中药和复方中药能降低TF的表达,机制是通过抑制TF mRNA、PKC和NF-κB表达,从而发挥抗血栓作用。
Tissue factor (TF) is a thrombus initiation factor under physiological coagulation and pathological conditions. It participates in the process of coagulation through interaction with factor VII, and plays an important role in thrombosis. To sum up the inhibitory effect of traditional Chinese medicine on tissue factor during thrombosis is of great significance for the development of anticoagulant and antithrombotic drugs targeting TF. The effects of single herbs and compound herbs on thrombus were tested by immunohistochemistry, enzyme linked immunosorbent assay (ELISA), reverse transcriptase polymerase chain reaction (RT-PCR), chromogenic substrate and western blot TF mRNA, PKC and NF-κB in the brain tissue were detected by Western blotting. The results showed that single Chinese herbal medicine and compound traditional Chinese medicine can reduce the expression of TF by inhibiting the expression of TF mRNA, PKC And NF-κB expression, which play an antithrombotic effect.
