为了探讨绝经后骨质疏松胶原代谢异常的分子机制 .采用RT PCR和Gel SDS 聚丙烯酰胺凝胶电泳(PAGE)分别检测卵巢切除大鼠骨组织Ⅰ型胶原mRNA水平及基质金属蛋白酶 (matrixmetalloproteinase,MMPs)的活性 ,免疫组化观察骨组织Ⅰ型胶原蛋白量 .卵巢切除大鼠骨组织Ⅰ型胶原mRNA表达降低约2 6 3% ,pro MMP 9活性明显提高 (P <0 0 5 ) ,应用雌激素治疗后Ⅰ型胶原mRNA表达较卵巢切除组增加34 1% ,pro MMP 9活性的增加明显降低 .卵巢切除后骨组织MMP 9明胶酶活性明显提高 ,应用雌激素后MMP 9明胶酶活性明显降低 (P <0 0 5 )表明骨Ⅰ型胶原mRNA表达减少和pro MMP 9活性升高是绝经后骨质疏松的机理之一
In order to investigate the molecular mechanism of abnormal collagen metabolism in postmenopausal osteoporosis rats, RT-PCR and Gel SDS polyacrylamide gel electrophoresis (PAGE) were used to detect the expression of collagen type Ⅰ mRNA and the expression of matrix metalloproteinase (MMPs) ), The expression of type Ⅰ collagen in bone tissue was observed by immunohistochemistry.The expression of type Ⅰ collagen mRNA was decreased by about 26.3% in ovariectomized rats and the activity of pro MMP 9 was significantly increased (P <0.05) Compared with ovariectomy group, the expression of type Ⅰ collagen mRNA increased by 34.1% and the pro MMP 9 activity decreased significantly after hormone treatment. The activity of MMP 9 gelatinase in bone tissue was significantly increased after ovariectomy. The MMP 9 gelatinase activity was significantly decreased after estrogen treatment (P <0.05) showed that decreased expression of type Ⅰ collagen mRNA and increased pro MMP 9 activity were one of the mechanisms of postmenopausal osteoporosis
