目的研究4种大黄多糖(A、B、C、D)以及其配伍体外抗轮状病毒(RV)作用。方法在体外用MTT法检测药物的毒性作用,从抗吸附、直接杀伤和抑制病毒增殖3方面检测4种大黄多糖抗RV的效果;并依中效原理,对大黄多糖A、C进行配伍,研究其体外抗病毒的作用,并探讨最适配伍比例。结果4种大黄多糖体外有预防RV感染和抑制病毒增殖作用。A、C以1∶1配伍后,对病毒的直接杀伤的合用效应与合用指数关系:当fa为≤0.68时,CI≤1,两药作用相加或协同。对病毒的增殖抑制的合用效应与合用指数关系:当fa为≤0.57时,CI≤1,两药作用相加或协同。结论4种大黄多糖体外具有较好地抗RV病毒作用,且大黄多糖A、C合用效果对病毒的直接杀伤作用和抑制病毒增殖作用优于其单独使用。
Objective To study the effects of four kinds of rhubarb polysaccharides (A, B, C, D) and their compatibility against rotavirus (RV) in vitro. Methods MTT assay was used to detect the toxic effects of the drugs. The anti-RV effects of four rhubarb polysaccharides were detected from the aspects of anti-adsorption, direct killing and inhibition of virus proliferation. According to the principle of neutral effect, the compatibility of rhubarb polysaccharides A and C was studied. Its antiviral effect in vitro, and explore the optimal compatibility ratio. Results Four kinds of rhubarb polysaccharide could prevent RV infection and inhibit virus proliferation in vitro. A, C 1: 1 compatibility, the direct combination of the killing effect of the virus and co-use index relationship: when fa is ≤ 0.68, CI ≤ 1, the two drugs add or synergistic effect. The combined effect of inhibition of virus proliferation and co-use index: when fa is ≤ 0.57, CI ≤ 1, the two drugs add or synergistic effect. Conclusion The polysaccharides of Rhubarb and Rhubarb can effectively inhibit RV virus in vitro, and the combination of Rhubarb polysaccharide A and C can kill the virus directly and inhibit the proliferation of the virus better than its single use.
