良好的高频再生能力是水稻叶绿体转化能否成功的重要因素。本研究选择最新选育的20个具有良好农艺性状的早熟水稻新品系为材料,分别剥取幼穗作为外植体,从基因型和培养基组成两个因素对水稻再生体系进行优化,筛选高频再生能力好的水稻品系。研究结果表明,水稻品系19的幼穗在含有2.0 mg/L2,4-D的MS培养基上愈伤诱导率为100%,经过连续8轮继代后在最适培养基SF3上仍能保持77.8%的分化率,且分化数多于10株的高频分化率为36.1%,具有作为叶绿体转化受体的潜力。
Good high-frequency regeneration ability is an important factor in the success of rice chloroplast transformation. In this study, 20 newly-developed early-maturing rice lines with the best agronomic traits were selected as the materials, and the young spikes were used as explants respectively. Two factors including genotype and medium were used to optimize the rice regeneration system, Good frequency of regeneration of rice lines. The results showed that the callus induction rate of rice line 19 in MS medium containing 2.0 mg / L 2,4-D was 100%, and remained on the optimum medium SF3 after 8 successive passages 77.8% differentiation rate, and the high frequency differentiation rate of more than 10 strains was 36.1%, which has the potential as a chloroplast transformation acceptor.
