以葡萄籽提取物为原料,研究用AB-8型大孔吸附树脂对原花青素分级纯化,建立了高效液相色谱(HPLC)方法,用此方法对葡萄籽提取物中原花青素定性、对低聚体(OPC)定量分析。为得到聚合度不同的原花青素,实验选择20%乙醇、40%乙醇、60%乙醇对原花青素进行梯度洗脱,洗脱液经真空干燥得原花青素固体,即洗脱液成品。HPLC分析结果显示:20%乙醇洗脱液中的原花青素是单体和OPC,OPC含量为17.42%;40%乙醇洗脱液中的原花青素是OPC和高聚体(OPC),OPC含量为80.32%;60%乙醇洗脱液中的原花青素是PPC。
Grape seed extract was used as raw material to study the purification of proanthocyanidins by AB-8 macroporous adsorption resin. HPLC method was established for the determination of proanthocyanidins in grape seed extract, (OPC) quantitative analysis. In order to obtain proanthocyanidins with different degrees of polymerization, the proanthocyanidins were selected with 20% ethanol, 40% ethanol and 60% ethanol for gradient elution. The eluent was dried under vacuum to obtain the proanthocyanidin solid, ie the eluate product. The results of HPLC showed that the proanthocyanidins in 20% ethanol eluent were monomers and OPC, the content of OPC was 17.42%. The proanthocyanidins in 40% ethanol eluent were OPC and high polymer (OPC) with OPC content of 80.32% The proanthocyanidin in the 60% ethanol eluent is PPC.
