目的 检测大鼠脑缺血再灌注后细胞外信号调节蛋白激酶信使RNA(ErkmRNA)的表达,探讨降钙素 基因相关肽(CGRP)和神经生长因子(NGF)对脑组织的保护作用。 方法 采用颈动脉负压分流法制作大鼠脑缺 血再灌注模型,采用原位杂交及显微图像分析方法检测海马及皮质内Erk的表达。 结果 大鼠缺血再灌注海马 及皮质内ErkmRNA较正常组增多(P<0.05),而注射CGRP或NGF后ErkmRNA明显高于缺血再灌注组(P< 0.01),两者联合应用效果更加显著(P<0.05)。 结论 CGRP及NGF可能参与缺血神经元ErkmRNA的调节,两 者对缺血神经元有协同修复作用。
Objective To detect the expression of extracellular signal-regulated kinase (ErkmRNA) after cerebral ischemia-reperfusion in rats and to explore the protective effect of calcitonin gene related peptide (CGRP) and nerve growth factor (NGF) on brain tissue. Methods Cerebral ischemia-reperfusion model was established by carotid artery negative pressure shunt method. The expression of Erk in hippocampus and cortex was detected by in situ hybridization and microscopic image analysis. Results Erk mRNA in the hippocampus and cortex of ischemia-reperfusion rats increased more than that of the normal group (P <0.05), while Erk mRNA expression of CGRP or NGF group was significantly higher than that of ischemia-reperfusion group (P <0.01) (P <0.05). Conclusion CGRP and NGF may be involved in the regulation of Erk mRNA in ischemic neurons. Both of them may play a synergistic role in the repair of ischemic neurons.
