目的检测信号转导子和转录激活子1(STAT1)在鼻咽癌中的表达,探讨其在鼻咽癌发生过程中的作用及其临床意义。方法采用SYBR GreenI荧光定量RT-PCR技术对48例鼻咽癌组织和12例慢性鼻咽炎组织中STAT1 mRNA的表达进行检测,并分析其表达情况与临床病理资料的相关性。结果鼻咽癌癌组织中STAT1 mRNA的表达水平明显高于慢性鼻咽炎组织(P<0.05);在肿瘤TNM分期中,T3+T4期癌组织STAT1 mRNA表达水平明显高于T1+T2期(P<0.05);在临床分期中,Ⅲ+Ⅳ期癌组织STAT1 mRNA表达水平明显高于Ⅰ+Ⅱ期(P<0.05);而其表达与有无淋巴结转移无明显相关(P>0.05)。结论 STAT1 mRNA在鼻咽癌组织中高表达,其表达增强可能与鼻咽癌的发生和发展相关,可作为鼻咽癌恶性潜能的检测指标之一。
Objective To detect the expression of signal transducer and activator of transcription 1 (STAT1) in nasopharyngeal carcinoma (NPC) and to explore the role of signal transducer and activator of transcription 1 (STAT1) in the pathogenesis of nasopharyngeal carcinoma and its clinical significance. Methods SYBR Green I fluorescence quantitative RT-PCR was used to detect the expression of STAT1 mRNA in 48 cases of nasopharyngeal carcinoma and 12 cases of chronic nasopharyngitis. The correlation between the expression of STAT1 mRNA and clinicopathological data was analyzed. Results The expression of STAT1 mRNA in nasopharyngeal carcinoma tissues was significantly higher than that in chronic nasopharyngitis tissues (P <0.05). The expression of STAT1 mRNA in stage T3 + T4 carcinomas was significantly higher than that in T1 + T2 stages <0.05). In the clinical stage, STAT1 mRNA expression in stage Ⅲ + Ⅳ cancer was significantly higher than that in stage Ⅰ + Ⅱ (P <0.05). However, the expression of STAT1 mRNA was not correlated with lymph node metastasis (P> 0.05). Conclusions STAT1 mRNA is highly expressed in nasopharyngeal carcinoma and its expression may be related to the occurrence and development of nasopharyngeal carcinoma and may be used as one of the detection indexes of malignant potential of nasopharyngeal carcinoma.
