目的从mRNA和蛋白质水平研究鸟氨酸脱羧酶在胃癌中的表达,探讨其在胃癌形成中的作用。方法采用半定量RT-PCR技术检测胃癌及相应癌旁组织中ODC mRNA的表达量,采用Western-blot在蛋白质水平检测胃癌及相应癌旁组织中ODC的表达量,并做统计学分析。结果21例胃癌组织中ODC在mRNA和蛋白质水平的表达量均高于癌旁组织(P<0.01)。ODC的表达量低分化胃癌组织显著高于高中分化(P<0.01)。结论ODC表达增强在胃癌发生发展中起重要作用,提示ODC可作为胃癌的基因诊断与治疗的靶点。
Objective To study the expression of ornithine decarboxylase in gastric carcinoma at mRNA and protein levels and to explore its role in gastric carcinogenesis. Methods ODC mRNA expression was detected by semi-quantitative RT-PCR in gastric cancer and its corresponding paracancerous tissues. Western-blot was used to detect the ODC expression in gastric cancer and corresponding paracancerous tissues at the protein level, and statistical analysis was made. Results The expression levels of ODC at 21 mRNA and protein levels in gastric cancer tissues were significantly higher than those in paracancerous tissues (P <0.01). ODC expression in poorly differentiated gastric cancer was significantly higher than that in high school (P <0.01). Conclusion The increased expression of ODC plays an important role in the development of gastric cancer, suggesting that ODC can be used as a target for gene diagnosis and treatment of gastric cancer.
