目的:探讨急性心肌梗死后梗死区周围心肌细胞内细胞周期蛋白A2(cyclin A2)的表达。方法:制作SD大鼠急性心肌梗死模型,分别于梗死后3d、1周、2周、3周、4周各处死5只大鼠,5只假手术大鼠为对照,取出心脏组织,进行Masson染色,同时使用免疫组织化学法检测心脏组织中cyclin A2及磷酸化组蛋白H3(phospho-histone H3,H3P)的表达。结果:Masson染色从病理上证明心肌梗死模型构建成功;梗死区周围心肌细胞cyclin A2的表达在梗死后1周时最高,阳性细胞染色百分比为(7.72±1.16)%,2周后逐渐下降,梗死后4周时,与假手术组相似,几乎无阳性表达。梗死后第3天、1周、2周和3周均可见少量心肌细胞核表达H3P,其阳性率为1.56%~3.12%。结论:心肌细胞cyclin A2的阳性表达提示急性心肌梗死后梗死区周围少部分心肌细胞能重新进入有丝分裂周期。
Objective: To investigate the expression of cyclin A2 in myocardial cells around infarct zone after acute myocardial infarction. Methods: Acute myocardial infarction (SD) model was made in SD rats. Five rats were sacrificed at 3d, 1, 2, 3 and 4 weeks after infarction. Five sham rats were used as controls. At the same time, the expression of cyclin A2 and phospho-histone H3 (H3P) in cardiac tissue were detected by immunohistochemical method. Results: The results of Masson staining showed that the model of myocardial infarction was established successfully. The expression of cyclin A2 in myocardial cells around the infarct area was the highest at 1 week after infarction. The percentage of staining positive cells was (7.72 ± 1.16)%, and gradually decreased after 2 weeks After 4 weeks, similar to sham group, almost no positive expression. On the 3rd day, 1 week, 2 weeks and 3 weeks after infarction, a small amount of myocardial cells expressed H3P, the positive rate was 1.56% -3.12%. Conclusion: The positive expression of cyclin A2 in cardiomyocytes suggests that a small number of myocardial cells around the infarct area may re-enter the mitotic cycle after acute myocardial infarction.
