目的:制作可应用于常压慢性间隙性缺氧(CIH)研究的动物模型,探讨CIH大鼠模型中氧化应激指标与主动脉内皮细胞损伤的关系。方法:24只SD大鼠随机分为空白对照组(正常饲养4周,UC组)、CIH组(CIH 4周)和复氧组(CIH 4周后复氧4周,RH组),每组8只。实验结束后测定大鼠血清中氧化应激指标丙二醛(MDA)、超氧歧化物(SOD)、NO的水平。处死大鼠取出腹主动脉,光镜下观察主动脉内皮细胞形态及内皮型一氧化氮合酶(eNOS)的表达情况。结果:CIH组SOD、NO较UC组均明显降低,而MDA显著升高(均P<0.05);大鼠腹主动脉内皮形态改变,eNOS表达减弱,明显低于UC组。RH组、UC组SOD、NO及MDA比较,差异无统计学意义(均P>0.05);2组大鼠腹主动脉内皮形态均未见明显改变,eNOS的表达比较差异无统计学意义(均P>0.05)。结论:大鼠CIH模型中氧化应激参与了主动脉的病变过程。
OBJECTIVE: To establish an animal model that can be applied to chronic hypoxia-induced hypoxia (CIH) in rats and to explore the relationship between oxidative stress and aortic endothelial cell injury in CIH rats. Methods: Twenty-four Sprague-Dawley rats were randomly divided into blank control group (4 weeks, UC group), CIH group (4 weeks) and reoxygenation group (4 hours after CIH reoxygenation group, RH group) . After the experiment, the level of oxidative stress indicators malondialdehyde (MDA), superoxide dismutase (SOD) and NO in serum were determined. The abdominal aorta was removed and the morphology of aortic endothelial cells and the expression of endothelial nitric oxide synthase (eNOS) were observed under light microscope. Results: The levels of SOD and NO in CIH group were significantly lower than those in UC group, while the levels of MDA in CIH group were significantly increased (all P <0.05). The endothelial morphology and the expression of eNOS in abdominal aorta of CIH group were significantly lower than those in UC group. There was no significant difference in the SOD, NO and MDA levels between RH, RH, and UC groups (all P> 0.05). There was no significant change in the endothelial morphology of abdominal aorta in both groups, and there was no significant difference in eNOS expression between the two groups P> 0.05). Conclusion: Oxidative stress is involved in the pathological changes of the aorta in rat CIH model.
