目的探讨基因的联合干扰对人胃癌SGC-7901细胞增殖的影响。方法将转染细胞分为共四组:psi-control对照组(A组)、psi-端粒酶催化亚基(hTERT)1组(B组)、psi-hTERT1和psi-hTERT2共转染组(C组)、psi-hTERT1和psi-端粒酶调节相关蛋白(TRAP)1共转染组(D组)。采用RT-PCR及Western blot分别检测hTERT mRNA和蛋白表达,端粒重复序列扩增-PCR法检测端粒酶活性,MTT法检测SGC-7901细胞增殖。结果与A组相比,B组、C组hTERT mRNA和蛋白表达、端粒酶活性降低(P<0.05),细胞增殖减慢,细胞凋亡增加;而D组各指标与A组比较差异无统计学意义(P>0.05)。结论联合干扰的效果可能与剂量和目的基因有关。
Objective To investigate the effect of gene interference on the proliferation of human gastric cancer cell line SGC-7901. Methods The transfected cells were divided into four groups: psi-control group (group A), psi- telomerase catalytic subunit group (group B), psi-hTERT1 and psi-hTERT2 co- (Group C), psi-hTERT1 and psi- telomerase regulatory-related protein (TRAP) 1 co-transfection group (group D). The expression of hTERT mRNA and protein was detected by RT-PCR and Western blot, respectively. The telomerase activity was detected by PCR amplification of telomere-repeats. The proliferation of SGC-7901 cells was detected by MTT assay. Results Compared with group A, the expression of hTERT mRNA and protein, the activity of telomerase in group B and group C were decreased (P <0.05), the cell proliferation was slowed down and the cell apoptosis was increased. The difference between group D and group A was no significant difference Statistical significance (P> 0.05). Conclusion The effect of combined interference may be related to dose and gene of interest.
