目的建立复方二神丸HPLC指纹图谱,并对该复方的两味组成药物补骨脂和肉豆蔻分别经过盐炙及煨炙后的复方二神丸HPLC指纹图谱进行比较研究,探讨经过炮制后的药物组成的复方二神丸在化学成分上的变化。方法采用HPLC色谱法;色谱柱Diamonsil C18柱色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-水梯度洗脱;检测波长254 nm;柱温30℃;流速1.0 ml/min。以指纹图谱软件计算相似度,对共有特征峰进行图谱比较。结果补骨脂、肉豆蔻生品同炮制品组方的二神丸HPLC指纹图谱中都标记出32个共有峰,未发现有成分的新增和消失,有4种成分在经过炮制药物的组方中含量明显降低,9种成分含量明显增高。结论该方法简便、快捷、重复性好,利用HPLC指纹图谱分析方法,可较全面的反映补骨脂、肉豆蔻炮制前后组方二神丸化学成分的差异。
OBJECTIVE To establish the HPLC fingerprint of Fufang Er Shen Pill, and to compare the HPLC fingerprints of Fufang Er Shen Pill after the salt-burning and the simmering respectively, Changes in the chemical composition of the drug Erzhu pill. Methods The column was separated on a Diamonsil C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase was acetonitrile-water gradient. The detection wavelength was 254 nm. The column temperature was 30 ℃ and the flow rate was 1.0 ml / min. Fingerprint software to calculate the similarity, the total characteristics of the peak spectrum comparison. RESULTS: There were 32 common peaks in the HPLC fingerprints of Er Shen pill in the prescriptions of psoralen and nutmeg, and no addition or disappearance of components was found. There were 4 components in the drug-treated group Fang significantly reduced the content of 9 components were significantly higher. Conclusion The method is simple, rapid and reproducible. By using HPLC fingerprinting method, the chemical composition of Fang-Er Shen-Pill can be more completely reflected before and after the preparation of psoralen and nutmeg.
