目的:研究血小板生成素(TPO)及其他造血生长因子在体外对人血小板活化的影响。方法:应用荧光标记单克隆抗体和流式细胞技术。结果:终浓度 120 ng/mL的TPO在体外能直接诱导人血小板活化,活化率为 8.89%~39.92%,中位数 17.43%,40ng/mL的较低浓度 TPO及 100ng/mL粒细胞-巨噬细胞集落刺激因子(GM-CSF)和白细胞介素-3(IL-3)对血小板活化无影响。结论:TPO对血小板功能有促进作用,较高浓度的TPO可直接诱导血小板活化。
Objective: To investigate the effect of thrombopoietin (TPO) and other hematopoietic growth factors on human platelet activation in vitro. Methods: Fluorescently labeled monoclonal antibodies and flow cytometry were used. Results: TPO with a final concentration of 120 ng / mL could directly induce human platelet activation in vitro with the activation rates of 8.89% -39.92%, median of 17.43%, lower concentrations of 40ng / mL TPO and 100ng / ML granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-3 (IL-3) had no effect on platelet activation. Conclusion: TPO can promote platelet function. Higher concentration of TPO can directly induce platelet activation.
