背景:脑缺血卒中的病理机制复杂,实施“鸡尾酒”方案可能是有前途的研究。目的:研究复方阿魏酸对实验性大鼠脑缺血再灌注损伤的保护作用,并探讨其作用机制。设计:完全随机设计、对照实验研究。地点和材料:本研究地点为重庆医科大学第二临床学院,健康成年Wis-tar大鼠由重庆医科大学实验动物中心提供。干预:采用大鼠双侧颈总动脉阻断和尾部放血并重复缺血再灌注的脑缺血模型,分别给予生理盐水、丹参药液或复方阿魏酸药液灌胃5d。主要观察指标:大鼠脑组织内皮素-1(Endothelin-1,ET-1)、降钙素基因相关肽(Calcitoningene-relatedpeptide,CGRP)、丙二醛、能量代谢、热休克蛋白70(heatshockprotein70,HSP70)表达及病理组织学的观察。结果:模型组ET-1、丙二醛含量最高,CGRP最低,存在能量代谢紊乱。复方阿魏酸高剂量组脑组织犤ET-1(0.73±0.04)ng/g犦、乳酸犤(16.28±1.1)μmol/g犦、丙二醛犤(116.8±7.55)nmol/g犦含量明显降低,ATP犤(18.22±0.83)μg/g犦,CGRP犤(1.62±0.13)ng/g犦、Na+-K+-ATP酶活性犤(747.82±63.94)nkat犦显著升高,与模型组相比,差异有显著性意义(P<0.05)。复方阿魏酸的以上作用存在量效关系,其高剂量组优于阳性对照药物丹参。复方阿魏酸治疗组大脑皮层HSP70表达明显增强,大脑缺血损伤程度?
Background: The pathological mechanism of cerebral ischemic stroke is complicated. The implementation of “cocktail” may be a promising research. Objective: To study the protective effect of compound ferulic acid on cerebral ischemia-reperfusion injury in rats and its mechanism. Design: Completely randomized design, controlled experimental study. Location and Materials: The study was conducted at the Second Clinical College of Chongqing Medical University. Healthy adult Wis-tar rats were provided by Experimental Animal Center of Chongqing Medical University. Intervention: The bilateral cerebral carotid artery occlusion and tail bleeding and repeated ischemia-reperfusion cerebral ischemia model were given saline, Salvia liquid or compound ferulic acid solution for 5 days. MAIN OUTCOME MEASURES: Endothelin-1 (ET-1), calcitonin gene-related peptide (CGRP), malondialdehyde, energy metabolism, heat shock protein 70 HSP70 expression and histopathological observation. Results: ET-1, MDA and CGRP in the model group were the highest, with the disturbance of energy metabolism.
