目的:建立衍生化GC-MS法同时测定不同来源南方红豆杉叶中19种脂肪酸甲酯化衍生物的含量。方法:采用HP-VOC(60m×320μm,1.8μm)弹性石英毛细管柱,程序升温:初温80℃,以10℃/min升至150℃保持5min,以3℃/min升至195℃保持5min,以2℃/min升至230℃保持10min,总计运行59.5min。进样口温度为250℃,分流比10∶1,进样量0.6μL,载气为氮气,流速为10m L/min。结果:19种脂肪酸的响应峰面积与其相应浓度的线性关系良好(r>0.9976)。加样回收率为94.85%-105.43%,RSD均小于4.0%。结论:该方法简便快速、准确,具有良好的重复性和回收率,可用于南方红豆杉叶中19种脂肪酸成分含量测定。不同来源的南方红豆杉叶中脂肪酸含量存在显著差异。
OBJECTIVE: To establish a derivatization GC-MS method for the simultaneous determination of 19 fatty acid methyl ester derivatives from different sources of Taxus chinensis. Methods: HP-VOC (60m × 320μm, 1.8μm) elastic quartz capillary column was used. The temperature was programmed as follows: the initial temperature was 80 ℃, the temperature was raised to 150 ℃ at 10 ℃ / min for 5min and the temperature was raised to 195 ℃ at 3 ℃ / , Held at 2 ° C / min to 230 ° C for 10 min, for a total run of 59.5 min. Inlet temperature was 250 ° C, split ratio 10: 1, injection volume 0.6 μL, carrier gas was nitrogen, flow rate 10m L / min. Results: The linear relationship between the peak areas of 19 fatty acids and their corresponding concentrations was good (r> 0.9976). The recovery rate was 94.85% -105.43%, RSD was less than 4.0%. Conclusion: The method is simple, rapid and accurate, and has good repeatability and recovery. It can be used to determine the content of 19 kinds of fatty acids in the leaves of Taxus chinensis. Different sources of Taxus chinensis leaves fatty acid content there are significant differences.
