目的探讨大鼠骨髓来源的间充质干细胞的培养和鉴定方法,为其功能研究奠定基础。方法采用全骨髓贴壁法进行骨髓来源的间充质干细胞的原代培养,通过酶消化法进行人工纯化,通过形态学观察、细胞周期测定及其自然分化实验进行鉴定。结果获得纯化的大鼠骨髓来源的间充质干细胞,该细胞呈长梭形或多角形,大小不等,生长密集,排列呈流线型。结论只要给细胞创造良好的生长条件并规范操作,全骨髓贴壁法可成功培养出稳定的大鼠骨髓来源的间充质干细胞。
Objective To explore the method of culturing and identifying rat bone marrow-derived mesenchymal stem cells, and to lay the foundation for its function study. Methods Primary bone marrow-derived mesenchymal stem cells were primary cultured with whole bone marrow adherent method and purified by enzymatic digestion. The morphological, cell cycle and natural differentiation assays were used to identify the bone marrow-derived mesenchymal stem cells. Results Purified rat bone marrow-derived mesenchymal stem cells were obtained. The cells were long fusiform or polygonal in size. They grew intensively and arranged in a streamlined pattern. Conclusion As long as the cells are given good growth conditions and regulate the operation, the whole bone marrow adherent method can successfully produce stable rat bone marrow-derived mesenchymal stem cells.
