[目的]研究不同条件下蝗虫(Locusta migratoria)节律基因pdp(Pyruvate dehydrogenase phosphatase)的mRNA表达水平,进一步揭示蝗虫节律的分子机制。[方法]通过中国科学院动物研究所内的转录组数据库中找出pdp基因的原型来设计引物,以群居型东亚飞蝗的头部的反转录cDNA为模板,克隆出pdp基因的全序列。把一天24小时等分成八个点,进行定时取样,以qRT-PCR技术进行不同时段pdp基因表达量的测定;并在不同处理条件下,测定pdp基因的表达量。[结果]节律基因pdp在蝗虫的不同处理条件下变化不大。[结论]该研究对了解飞蝗节律规律﹑种族特点及有效防虫具有重要的现实意义。
[Objective] The research aimed to study the mRNA expression of pdp (Locusta migratoria) rhythm gene pdp under different conditions and further reveal the molecular mechanism of locust rhythm. [Method] Primers were designed by finding out the prototype of pdp gene in transcriptome database of Institute of Zoology, Chinese Academy of Sciences. The full-length sequence of pdp gene was cloned by reverse transcriptase cDNA from the head of gregarious migratory locusts. The 24-hour day was divided into eight points for timing sampling, and qRT-PCR was used to determine the expression of pdp gene at different time points. The expression of pdp gene was determined under different treatment conditions. [Result] The rhythm gene pdp did not change much under different treatments of locusts. [Conclusion] The study has important practical significance for understanding rhythm, race characteristics and effective pest control of locusts.
