从克山病病区粮和非病区粮饲养大鼠心肌中提取总 RNA,分析其中的α和β—CMHCmRNA 含量的变化。结果发现,与非病区粮组相比,病区粮组大鼠心肌中的α—CMHC m-RNA 含量逐渐降低,而β—CMHC mRNA 含量逐渐升高。在大鼠心肌 CM—ATP 酶活性的分析中发现,病区粮组大鼠 CM—ATP 酶活性的降低与 CM 基因表达变化之间具有良好的相关性。从而证明 CM—ATP 酶活性的变化是由于α—CMHC 基因表达减少和β—CMHC 基因表达增多,进而导致 CM 同工酶由 V_1向 V_3转化的结果。
Total RNA was extracted from the myocardium of rats fed on food and non-wedged grain from Keshan disease group, and the changes of α and β-CMHC mRNA content were analyzed. The results showed that, compared with the non-ward food group, ward rats in the ward group myocardial α-CMHC m-RNA content decreased gradually, and β-CMHC mRNA content gradually increased. In the analysis of myocardial CM-ATPase activity in rats, it was found that there was a good correlation between the decrease of CM-ATPase activity and the change of CM gene expression in wistar rats. Thus, the change of CM-ATPase activity is attributed to the decrease of α-CMHC gene expression and the increase of β-CMHC gene expression, leading to the conversion of CM isozyme from V_1 to V_3.
