为了从酶、基因水平阐明甾醇及三萜的生物合成机制,以人参毛状根进行了研究。曾报告由人参毛状根制备的微粒体组分中,检测到由氧化角鲨烯向环阿尔廷醇、达玛烯二醇及β-香树脂醇环化的3种酶活性。本次进行了环阿尔廷醇合酶的cDNA克隆。 由皂甙产量最多的培养第3周的人参毛状根提取mRNA,以逆转录得到的cDNA为模板,用在氧化角鲨烯环化酶间同源性非常高的序列(拟南芥第162、467、556及623开始的序列)设定的4种引物进行嵌套式
