目的探讨胎盘组织匀浆上清对小鼠骨髓树突状细胞生物学特征的影响。方法分离BALB/c小鼠骨髓单个核细胞,诱导分化为树突状细胞,经胎盘组织匀浆上清作用后,流式细胞仪检测树突状细胞的表型和抗原摄取能力,MTT法检测树突状细胞刺激同种异基因淋巴细胞反应的能力,ELISA法检测其分泌的细胞因子水平。结果正常对照小鼠和妊娠小鼠的骨髓树突状细胞的生物学特征没有明显差异(P≥0.05),而经胎盘组织匀浆上清作用后的正常小鼠及妊娠小鼠的CD11b+树突状细胞(MDC)明显降低,CD8α+树突状细胞(LDC)明显增高(P≤0.01),胎盘组织匀浆上清作用后的树突状细胞的抗原摄取能力及其刺激同种异基因淋巴细胞反应的能力明显低于对照(P≤0.01),可以分泌较高水平的IL-10和较低水平的IL-12,与对照相比较,P≤0.01。结论胎盘组织匀浆上清作用后的骨髓树突状细胞的生物学特征的改变可能与妊娠免疫耐受有关。
Objective To investigate the effect of placenta tissue homogenate on the biological characteristics of mouse bone marrow dendritic cells. Methods Bone marrow mononuclear cells of BALB / c mice were isolated and induced to differentiate into dendritic cells. The placental homogenate supernatant was used to detect the phenotype and antigen uptake ability of dendritic cells by flow cytometry. MTT assay The ability of dendritic cells to stimulate allogeneic lymphocyte reaction and the secretion of cytokines by ELISA assay. Results The biological characteristics of dendritic cells in normal control and pregnant mice showed no significant difference (P> 0.05), while CD11b + dendrites in normal mice and pregnant mice after placental homogenate supernatant (MDC), CD8α + dendritic cells (LDC) were significantly increased (P≤0.01), the antigen uptake capacity of dendritic cells after placental homogenate supernatant and its ability to stimulate allogeneic lymph node The cellular response was significantly lower (P <0.01) than the control (P <0.01), secreting higher levels of IL-10 and lower levels of IL-12 compared to controls, P <0.01. Conclusion The changes of biological characteristics of myeloid dendritic cells after placental homogenate supernatant may be related to immune tolerance in pregnancy.
