目的探讨杏仁核电点燃及海人酸点燃难治性癫大鼠模型脑组织及外周血P糖蛋白(Pgp)过度表达的可能机制,并比较不同方法致难治性癫大鼠脑组织及外周血中Pgp的表达趋势。方法应用电刺激点燃及海人酸点燃方法对大鼠杏仁核进行点燃,制作难治性癫模型;将其分为A组(杏仁核电点燃,12只),B组(杏仁核海人酸点燃,12只),另设C组(杏仁核埋入电极而不点燃,且给予卡马西平灌胃,8只),D组(杏仁核注射生理盐水,且给予卡马西平灌胃,8只),E组(杏仁核埋入电极而不点燃,且给予生理盐水灌胃,8只),F组(杏仁核注射生理盐水,且给予生理盐水灌胃,8只);采用免疫组化方法分析两种模型大鼠脑组织及外周血Pgp的表达。结果A组、B组大鼠脑组织及外周血Pgp表达明显高于其余4组,差异有统计学意义(均P<0.05);C组、D组大鼠脑组织及外周血Pgp表达高于E组及F组,差异有统计学意义(均P<0.05);A组与B组大鼠脑组织及外周血Pgp的表达差异无统计学意义。结论脑组织及外周血Pgp的过度表达可能是未能控制的癫发作、服用抗癫药(AEDs)等多因素共同作用的结果;难治性癫发生1个月后,脑组织中Pgp高表达的大鼠外周血中Pgp的表达同样高于脑组织中Pgp低表达大鼠外周血Pgp的表达,提示难治性癫大鼠外周血及脑组织中Pgp的表达趋势具有一定的相关性。
Objective To investigate the possible mechanism of overexpression of Pgp in brain tissue and peripheral blood of amygdala and kainic acid-refractory epilepsy rats and to compare the effects of different methods on brain tissue and Pgp expression in peripheral blood. Methods The model of refractory epilepsy was prepared by lighting the amygdala with electrical stimulation and kainic acid ignition. The rats were divided into group A (amygdala, 12), group B (amygdala 12 rats in group A), another group C (amygdalus was buried in the electrode without ignition, and carbamazepine was given into the stomach, 8 rats), Group D (saline injected with amygdala and carbamazepine, Only group A, group E, the amygdala was embedded in the electrode without igniting, and the rats were given normal saline gavage, 8 rats in each group. Group F (normal saline injected with saline of the amygdala and 8 rats) Methods The expression of Pgp in brain tissue and peripheral blood of two models of rats was analyzed. Results The expression of Pgp in brain tissues and peripheral blood of group A and group B was significantly higher than that of the other four groups (all P <0.05). The expression of Pgp in brain tissue and peripheral blood of group C and D was higher than that of group C E group and F group, the difference was statistically significant (all P <0.05). The expression of Pgp in brain tissue and peripheral blood of group A and group B had no statistical significance. Conclusion The overexpression of Pgp in brain tissue and peripheral blood may be the result of uncontrolled epileptic seizures and antiepileptic drugs (AEDs) and other factors. After 1 month of intractable epilepsy, brain tissue The expression of Pgp in Pgp-overexpressing rat peripheral blood was also higher than that in Pgp-overexpressing rat brain tissue, suggesting that Pgp expression in peripheral blood and brain tissue of refractory epileptic rats had a certain Correlation.
