目的通过研究土槿乙酸衍生物土槿乙酸酰胺(PB-LY)对HeLa细胞周期的影响,探讨PB-LY的抗肿瘤作用机制。方法采用流式细胞术检测PB-LY对HeLa细胞周期的影响;采用RT-PCR法检测PB-LY对He-La细胞周期相关基因p53和p21mRNA表达的影响。结果PB-LY可明显减少G1期的HeLa细胞比例,明显增加G2+M期细胞的比例,上调p53基因的表达,同时协同下调p21基因的表达。结论PB-LY可通过增加细胞周期相关基因p53基因的表达和减少p21基因的表达,使HeLa细胞的周期阻断在G2+M期,从而阻滞HeLa细胞的生长。
Objective To study the effect of PB-LY on the cell cycle of HeLa cells and explore the antitumor mechanism of PB-LY. Methods Flow cytometry was used to detect the effect of PB-LY on the cell cycle of HeLa cells. RT-PCR was used to detect the effect of PB-LY on the expression of p53 and p21 mRNA of He-La cell cycle-related genes. Results PB-LY can significantly reduce the proportion of HeLa cells in G1 phase, significantly increase the proportion of G2+M phase cells, up-regulate the expression of p53 gene, and simultaneously down-regulate the expression of p21 gene. Conclusion PB-LY can block the growth of HeLa cells by increasing the expression of p53 gene and decreasing the expression of p21 gene.
