目的:观察三七总皂甙预处理对心肌缺血再灌注大鼠血清肿瘤坏死因子-α的影响。方法:采用开胸冠状动脉结扎建立缺血再灌注模型,25只雄性SD大鼠随机分为假手术组(n=4)、缺血再灌注组(IR组,n=7)及缺血预处理组(IPC组,n=7)及三七总皂甙预处理组(PNS预处理组,n=7)。分别于再灌注后2h测定各组血清肌酸磷酸激酶-MB(CK-MB)含量以及肿瘤坏死因子-α(TNF-α)。结果:PNS预处理组血清CK-MB含量为5098.43±1268.18U/L,明显低于IR组(17925.71±2022.74U/L,P(0.05);但高于IPC组(2304.29±540.04U/L,P<0.05)。IPC组及PNS预处理组血清TNF-α含量分别为1.08±0.08ng/ml、1.11±0.18ng/nl,均明显低于IR组(1.51±0.24ng/ml,两组均P<0.05);两组间无显著差异(P>0.05)。结论:PNS预处理早期通过抑制TNF-α释放对缺血再灌注心肌产生早期保护作用,但其保护作用不及缺血预处理。
OBJECTIVE: To observe the effect of panax notoginseng saponin pretreatment on serum TNF-α in rats with myocardial ischemia-reperfusion injury. METHODS: A rat model of ischemia-reperfusion was established by thoracotomy and 25 male SD rats were randomly divided into sham operation group (n=4), ischemia-reperfusion group (IR group, n=7) and ischemic preconditioning. Treatment group (IPC group, n=7) and panax notoginseng pretreatment group (PNS pretreatment group, n=7). Serum creatine phosphokinase-MB (CK-MB) levels and tumor necrosis factor-α (TNF-α) levels were measured at 2 h after reperfusion. Results: The serum CK-MB level in the PNS pretreatment group was 5089.43±1268.18 U/L, which was significantly lower than that of the IR group (17925.71±2022.74 U/L, P(0.05); but higher than the IPC group (2304.29±540.04 U/L, P<0.05).The serum levels of TNF-α in the IPC and PNS pretreatment groups were 1.08±0.08ng/ml and 1.11±0.18ng/nl, respectively, which were significantly lower than those in the IR group (1.51±0.24ng/ml). P<0.05); There was no significant difference between the two groups (P>0.05).Conclusion: Early protection of TNF-α by PNS pretreatment has an early protective effect on ischemic reperfusion of myocardium, but its protective effect is less than that of ischemic preconditioning.
