目的:观察复方血栓通胶囊对糖尿病大鼠周围神经病变的疗效并探讨其作用机制。方法:选取SD大鼠32只,禁食12h,一次性腹腔注射链脲佐菌素(65mg/kg)诱导糖尿病模型,造模后将大鼠随机分为模型组和复方血栓通胶囊组。另选取11只大鼠做为正常对照组。每组每天干预1次,12周后检测大鼠的热痛潜伏期、神经血流量、微循环血流灌注量及神经传导速度,并观察坐骨神经病理变化。结果:与正常组比较,模型组大鼠热痛潜伏期延长,神经血流量减少,神经传导速度减慢;髓鞘松解;耳部及上肢末端微循环血流灌注量明显减少;下肢末端血流灌注量减少。与模型组比较,复方血栓通胶囊组大鼠热痛潜伏期缩短,神经血流量增大,神经传导速度增快,神经束膜整齐,耳部及上肢末端微循环血流灌注量明显增加,下肢末端血流灌注量增加。结论:复方血栓通可能是通过增加周围神经组织的微循环灌注量,对糖尿病大鼠周围神经病变进行改善。
Objective: To observe the curative effect of compound Xueshantong capsule on peripheral neuropathy in diabetic rats and to explore its mechanism. Methods: Twenty-two SD rats were selected and fasted for 12h. Diabetic rats were induced by intraperitoneal injection of streptozotocin (65mg / kg). The rats were randomly divided into model group and compound Xueshuantong capsule group. Another eleven rats as a normal control group. The rats in each group were intervened once a day. After 12 weeks, the thermal latency, nerve blood flow, microcirculation perfusion and nerve conduction velocity in rats were measured and the pathological changes of sciatic nerve were observed. Results: Compared with the normal group, the latent period of thermal pain in model group was prolonged, the nerve blood flow decreased, the nerve conduction velocity was slowed down, the myelin sheath was loosened, the amount of microcirculation perfusion in ear and upper extremity was significantly reduced, Decreased perfusion. Compared with the model group, the rats in Fufang Xueshuantong Capsule group had shorter heat stroke latency, increased nerve blood flow, faster nerve conduction velocity, neuraxial tunica intima, increased microcirculation perfusion in the ear and upper extremity, Increased blood perfusion. Conclusion: Compound Xueshuantong may improve peripheral neuropathy in diabetic rats by increasing microcirculation perfusion of peripheral nerve tissue.
