目的研究亚慢性低剂量2,3,7,8-四氯二苯-并-二噁英(2,3,7,8-tetrachlorodibenzo-p-dioxin,TCDD)染毒对SD雄性成年大鼠海马形态结构的影响。方法将32只1月龄SPF级Sprague-Dawley雄性大鼠随机分为4组,分别为对照(玉米油)组和低(2 ng/kg)、中(10 ng/kg)、高剂量(50 ng/kg)TCDD染毒组,每组8只。将相应体积的TCDD染毒溶液(或玉米油)涂抹在面包片(与动物标准饲料成分相当,排除二噁英污染)上,供动物自由食用,每天1次,连续染毒13周。取海马组织进行电镜超微结构观察,常规HE染色,海马CA1区体视学分析。结果 TCDD染毒大鼠海马各区细胞排列较对照组松散,锥体细胞超微结构的损伤以内质网同心圆样改变和线粒体空泡样变为主。各剂量TCDD染毒雄性大鼠海马CA1区锥体细胞的体积密度、表面积密度、平均体积、比表面积与对照组相比,差异均无统计学意义。结论亚慢性低剂量TCDD染毒可引起成年雄性大鼠海马区细胞排列松散,CA1区锥体细胞超微结构受到破坏。
Objective To investigate the effects of sub-chronic low-dose 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) exposure on SD male adult hippocampus Effects of morphological structure. Methods Thirty-two male SPF Sprague-Dawley rats aged 1 month were randomly divided into 4 groups: control (corn oil) group and low (2 ng / kg) ng / kg) TCDD exposure group, 8 rats in each group. Results The arrangement of cells in the hippocampus of TCDD-exposed rats was looser than that in the control group. The ultrastructural damage of pyramidal cells was mainly concentric with endoplasmic reticulum and mitochondria-like vacuolization. The volume density, surface area density, average volume and specific surface area of pyramidal cells in hippocampal CA1 of TCDD-treated male rats were not significantly different from those of control group. Conclusion Subchronic low dose TCDD can cause loosely arranged cells in the hippocampus of adult male rats and disrupt the ultrastructure of pyramidal cells in CA1 area.
