[目的]了解鞘内注射PI3K特异性抑制剂LY294002对小鼠股骨癌痛的影响。[方法]C3H/HeJ小鼠32只,随机分为对照组、假手术组、骨癌组和鞘内LY294002+骨癌组(抑制剂组),每组8只,分别于造模前和模型建立后的第8、10、12、14、17、21d测定各小鼠机械痛敏(机械缩腿阈值,mechanical withdrawal threshold,MWT)和热痛敏(热缩腿潜伏期,thermal withdrawal latency,TWL),LY294002术前鞘内注射。[结果]与术前相比较,骨癌组小鼠术后12、14、17、21dMWT明显下降(t=4.527,P=0.003;t=3.698,P=0.008;t=4.043,P=0.005;t=4.309,P=0.004),TWL明显降低(t=5.946,P=0.001;t=5.214,P=0.001;t=6.786,P=0.000;t=5.758,P=0.001);与假手术组相比较,骨癌组小鼠术后12、14、17、21dMWT明显下降(P=0.027;P=0.000;P=0.000;P=0.000),TWL明显降低(P=0.000;P=0.000;P=0.000;P=0.000)。与骨癌组相比较,抑制剂组小鼠在12、14、17、21dMWT明显升高(P=0.031;P=0.001;P=0.002;P=0.000),TWL明显延长(P=0.002;P=0.000;P=0.000;P=0.000);与术前和对照组相比较,假手术组在各时间点的MWT和TWL均无明显变化(P>0.05)。[结论]鞘内注射PI3K特异性抑制剂LY294002能明显减轻骨癌痛小鼠机械痛敏和热痛敏,提示PI3K在脊髓水平参与了骨癌痛信息传递。
[Objective] To investigate the effect of intrathecal PI3K-specific inhibitor LY294002 on femur cancer pain in mice. [Methods] Thirty-two C3H / HeJ mice were randomly divided into control group, sham operation group, bone cancer group and intrathecal LY294002 + bone cancer group (inhibitor group), with 8 rats in each group. The mechanical pain thresholds (mechanical withdrawal threshold, MWT) and thermal withdrawal latency (TWL) were measured on the 8th, 10th, 12th, 14th, LY294002 intrathecal injection. [Results] The MWT of 12, 14, 17 and 21 days after operation in bone cancer group was significantly lower than that before operation (t = 4.527, P = 0.003, t = 3.698, (t = 5.946, P = 0.001; t = 5.214, P = 0.001; t = 6.786, P = 0.000; t = 5.758, P = 0.001; t = 4.309, P = 0.004) Compared with the control group, the BWT decreased significantly at 12, 14, 17 and 21 days after operation (P = 0.027; P = 0.000; P = 0.000; P = 0.000) = 0.000; P = 0.000). Compared with the bone cancer group, the TWT was significantly prolonged at 12, 14, 17 and 21 dMWT in the inhibitor group (P = 0.0031; P = 0.001; P = 0.002; P = = 0.000; P = 0.000; P = 0.000). Compared with preoperative and control groups, MWT and TWL did not change significantly in sham operation group at each time point (P> 0.05). [Conclusion] Intrathecal injection of PI3K-specific inhibitor LY294002 can significantly reduce the mechanical pain and thermal hyperalgesia in bone cancer pain mice, suggesting that PI3K is involved in the transmission of bone cancer pain information at spinal level.
