目的构建百草枯急性中毒致家兔肺纤维化模型。方法以50mg/kg的20%百草枯水溶液一次灌胃构建百草枯致家兔肺间质纤维化模型。于染毒前和染毒后1、3、7d通过耳缘静脉采血进行Hyp活性测定。于染毒后3、7、14、21d分批处死家兔,观察肺组织外观并收集肺组织进行光镜检查。结果染毒组和对照组家兔血Hyp活性组间差异有统计学意义,且两组间血Hyp活性在染毒后不同时相变化趋势不同,染毒组家兔高于对照组(P<0.05)。肺组织病理检查显示,染毒组家兔染毒后21d出现明显的纤维化改变。结论以50mg/kg 20%百草枯水溶液一次给予家兔灌胃能够在染毒后3周发生肺纤维化。
Objective To construct a model of pulmonary fibrosis in rabbits with acute paraquat poisoning. Methods A rat model of paraquat-induced pulmonary fibrosis was established by intragastric administration of 50mg / kg 20% paraquat aqueous solution. Hyp activity was measured by blood sampling through the ear vein before and 3, 7 days after exposure. Rabbits were killed in batches 3, 7, 14 and 21 days after exposure, the appearance of lung tissue was observed and lung tissue was collected for light microscopy. Results There was a significant difference in Hyp activity between the two groups (P <0.05). The Hyp activity of Hyp in the two groups was different at different time points after exposure, and that in the control group was significantly higher than that in the control group (P < 0.05). Pathological examination of lung tissue showed that the exposed group showed obvious fibrosis 21 days after exposure. Conclusions The intragastric administration of 50mg / kg 20% paraquat aqueous solution to rabbits can cause pulmonary fibrosis 3 weeks after exposure.
