目的通过研究活血通络汤对激素性股骨头坏死(steroid induced avascular necrosis of femoral head,SANFH)模型中VEGF(vascular endothelial growth factor,血管内皮因子)、TNF-α(Tumor necrosis factor,肿瘤坏死因子α)的表达变化,来阐述活血通络汤对激素性股骨头坏死的作用机制。方法将192只日本大耳兔随机分组,模型组A、模型组B、模型对照组C、空白对照组D各48只,A、B、C组运用贺西京造模法造模。A组第2周开始喂服中药饲料,B组第3周开始喂服中药饲料,C、D组均喂服等量普通饲料,并于第2、5、8周末检测血中VEGF、TNF-α的含量,观察股骨头病理,计算空骨陷窝率。结果在不同时期比较,随实验时间的延长,在骨陷窝率方面,A组有下降趋势,B、C组均逐渐上升,但B组上升幅度小于C组;在血清VEGF方面,C组呈逐渐下降趋势,A组呈上升趋势,B组虽然呈下降趋势,但其下降幅度要明显小于C组;在血清TNF-α方面,C组逐渐上升,A、B组血清中TNF-α呈现下降趋势(第5周有升高)。结论激素可以导致血中VEGF的表达减弱,TNF-α的表达增强,活血通络汤具有抑制激素使VEGF表达减弱的趋势,下调TNF-α,从而缓解股骨头坏死进展,促进股骨头坏死的修复。
Objective To study the effect of Huoxuetongluo Decoction on vascular endothelial growth factor (VEGF), tumor necrosis factor-α (TNF-α) in steroid induced avascular necrosis of femoral head (SANFH) ) To expound the mechanism of action of Huoxue Tongluo Decoction on steroid-induced femoral head necrosis. Methods A total of 192 Japanese large-eared rabbits were randomly divided into three groups: model group A, model group B, model control group C, and blank control group D, 48 animals each. Groups A, B and C were made by He Xijing modeling. The rats in group A started to receive traditional Chinese medicine feed from the second week, the traditional Chinese medicine feed was fed from the third week in group B, the same amount of common feed in groups C and D, and the levels of VEGF, TNF- α content, observe the pathology of femoral head, calculate the rate of empty lacuna. Results In different periods, with the extension of experimental time, there was a downward trend in the rate of bone lacunar in group A, while the level in group B and C increased gradually, but the increase in group B was less than that in group C. In terms of serum VEGF, group C was In group A, B and C showed a decreasing trend, while the decreasing trend in group B was significantly less than that in group C. In serum TNF-α, group C gradually increased, while group A and B showed a decrease in serum TNF-α Trend (Week 5 increased). Conclusion Hormone can decrease the expression of VEGF and increase the expression of TNF-α. Huoxue Tongluo Decoction can inhibit the expression of VEGF and decrease the expression of TNF-α, so as to relieve the progression of femoral head necrosis and promote the repair of femoral head necrosis .
