目的:建立芒果苷的高效液相色谱测定法,研究其在Caco-2细胞上的转运特征。方法:利用MTT实验筛选出芒果苷在Caco-2细胞中的安全浓度,然后用Caco-2细胞单层模型研究芒果苷的转运特征。采用HPLC法检测药物浓度,考察时间、药物浓度、P-糖蛋白抑制剂对芒果苷在Caco-2细胞上转运的影响。结果:芒果苷在Caco-2细胞单层膜上的转运量具有一定的浓度和时间依赖性,P-糖蛋白抑制剂维拉帕米和环孢素A能够增加芒果苷跨膜转运量。结论:芒果苷在Caco-2细胞上转运表现为被动扩散且有外转运蛋白参与芒果苷跨膜转运。
OBJECTIVE: To establish a method for the determination of mangiferin by HPLC and to study its transport characteristics on Caco-2 cells. Methods: The safe concentration of mangiferin in Caco-2 cells was screened by MTT assay, and the transport characteristics of mangiferin were studied by Caco-2 cell monolayer model. The drug concentration was determined by HPLC. The effects of time, drug concentration and P-glycoprotein inhibitor on the transport of mangiferin on Caco-2 cells were investigated. Results: The transport of mangiferin on monolayers of Caco-2 cells was dose-dependent and time-dependent. P-glycoprotein inhibitors verapamil and cyclosporin A could increase the transmembrane transport of mangiferin. CONCLUSION: Mangiferin translocates on Caco-2 cells with passive diffusion and transporters involved in transmembrane transport of mangiferin.
