目的:探讨中药复方肝欣液体外抗乙肝病毒(HBV)活性及抑制乙肝表面抗原(HBsAg)的作用。方法:采用2.2.15细胞培养测定肝欣液对HBV的活性以及采用反相被动血凝抑制试验(RPHI)对肝欣液抗HBsAg进行实验观察。结果:肝欣液最高浓度为5~7μl/ml,作用7天和14天时,对2.2.15细胞HBsAg分泌抑制率分别为55.12%~57.48%和60.98%~71.34%,并可使培养上清中HBVDNA的量减少;浓度为7μl/ml,作用14天时对乙肝e抗原(HBeAg)分泌抑制率为50.28%,并可使培养上清HBVDNA的量减少,肝欣液500μl/ml对HBsAg有显著抑制作用,与阴性对照组比较凝集效价下降3~4个倍比度(P<0.01)。结论:肝欣液浓度为5~7μl/ml时,在体外细胞培养中,具有一定的抗HBV活性,并对HBsAg有良好的抑制作用。
Objective: To investigate the anti-hepatitis B virus (HBV) activity and the inhibitory effect of hepatitis B surface antigen (HBsAg) in vitro. Methods: 2.2.15 cell culture was used to determine the activity of Gansu liquid on HBV and the anti-HBsAg activity of Ganshen liquid was observed by reverse phase hemagglutination inhibition test (RPHI). Results: The maximum inhibitory rates of HB-Ag in 2.2.15 cells were 55.12% -57.48% and 60.98% -71.34 %, And can reduce the amount of HBVDNA in culture supernatant; the concentration of 7μl / ml, the effect of 14 days on the secretion of hepatitis B e antigen (HBeAg) inhibition was 50.28%, and the amount of HBVDNA can reduce the culture supernatant, Hepatexin 500μl / ml had a significant inhibitory effect on HBsAg, and the agglutination titer decreased by 3 ~ 4 times (P <0.01) compared with the negative control group. Conclusion: When the concentration of hepatic extract is 5 ~ 7μl / ml, it has certain anti-HBV activity in in vitro cell culture and has a good inhibitory effect on HBsAg.
