目的探讨基质金属蛋白酶1(MMP-1)和金属蛋白酶1组织抑制因子(TI MP-1)基因与胃癌临床生物学行为的关系。方法采用逆转录-聚合酶链反应(RT-PCR)检测60例胃癌组织及其相应癌旁正常胃黏膜组织中MMP-1和TI MP-1 mRNA的表达。结果60例胃癌组织中MMP-1表达水平明显高于癌旁正常组织,TI MP-1表达水平明显低于癌旁正常组织(P<0.05)。胃癌组织中,MMP-1 mRNA的相对表达强度、未分化癌明显高于高、中分化癌,Ⅲ~Ⅳ期明显高于Ⅰ~Ⅱ期,伴淋巴结转移者明显高于无淋巴结转移者(P<0.05);TI MP-1 mRNA的相对表达强度低、未分化癌则明显低于高、中分化癌,Ⅲ~Ⅳ期明显低于Ⅰ~Ⅱ期,伴淋巴结转移者明显低于无淋巴结转移者(P<0.05)。但MMP-1、TI MP-1表达与患者年龄、性别、肿瘤病理类型及肿瘤大小和部位等临床参数无关(P>0.05)。MMP-I与TI MP-1表达呈负相关(r=-0.513,P<0.05)。结论胃癌中MMP-1、TI MP-1表达与胃癌分化程度、TNM分期及淋巴结转移有关。
Objective To investigate the relationship between the expression of matrix metalloproteinase 1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TI MP-1) and the clinical biological behavior of gastric cancer. Methods The expressions of MMP-1 and TI-MP-1 mRNA in 60 cases of gastric cancer and their adjacent normal gastric mucosa tissues were detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The expression of MMP-1 in 60 cases of gastric cancer tissues was significantly higher than that in adjacent normal tissues, and the expression of TI MP-1 was significantly lower than that in adjacent normal tissues (P <0.05). The relative intensity of MMP-1 mRNA expression in undifferentiated carcinoma was significantly higher than that in high-grade and moderately differentiated carcinoma in stage Ⅲ-Ⅳ, and was significantly higher than that in stage Ⅰ-Ⅱ in patients with lymph node metastasis <0.05). The relative expression intensity of TI-MP-1 mRNA was lower in undifferentiated carcinoma than that in high-differentiated and moderately-differentiated carcinoma. The expression of TI-MP-1 mRNA in stage Ⅲ-Ⅳ was significantly lower than that in stage Ⅰ-Ⅱ. (P <0.05). However, the expression of MMP-1 and TI-MP-1 had no relation with clinical parameters such as age, sex, tumor pathological type, tumor size and location (P> 0.05). The expression of MMP-1 and TI MP-1 was negatively correlated (r = -0.513, P <0.05). Conclusion The expression of MMP-1 and TI-1 in gastric cancer is related to the differentiation of gastric cancer, TNM stage and lymph node metastasis.
