人脐带血与骨髓、外周血干祖细胞生物学特性虽基本相同,但同时也存在一些不同的特性。通过造血祖细胞体外半固体培养发现3种不同来源的粒系祖细胞(CFU—GM)体外生长所需条件不同。骨髓和外周血CFU—GM培养需要外源细胞生长因子GM—CSF存在。脐血CFU—GM培养仅需要脐血浆单独存在即可获得较高的集落产率。揭示了脐血浆中存在一种脐血祖细胞依赖的成分。它不同于GM—CSF,也可能不是GM—CSF协同因子,尚待进一步探讨。此外,提出了一个脐血粒系祖细胞体外培养简便易行的培养体系。
Human umbilical cord blood and bone marrow, peripheral blood stem and progenitor cells are basically the same biological characteristics, but there are also some different characteristics. In vitro semi-solid culture of hematopoietic progenitor cells revealed different conditions for the in vitro growth of three different derived myeloid progenitor cells (CFU-GMs). Cultures of bone marrow and peripheral blood CFU-GM require the presence of exogenous cell growth factor GM-CSF. Umbilical blood CFU-GM culture requires only umbilical plasma alone to obtain higher colony yields. Revealed a umbilical cord blood cord blood progenitor cell-dependent components. It is different from GM-CSF, may not be synergistic factor GM-CSF, remains to be further explored. In addition, an umbilical cord blood progenitor cells cultured in vitro is simple and easy to culture system.
