目的:探讨脾阳虚模型大鼠“运”“化”功能的增龄性变化。方法:将新出生40只Wistar大鼠,体质量(5.6±0.5)g,采用肩胛骨间棕色脂肪组织切除术及高脂饮食隔日寒冷环境刺激造模,术后随机分为8周龄模型组、16周龄模型组、32周龄模型组、64周龄模型组4组,每组10只。另外40只大鼠用来设立相对应的同周龄的对照组4组,每组10只。到了相应的周龄后,检测胃肠动力学指标,采用免疫组化检测胃促生长素表达,酶联免疫吸附法和放射免疫法检测血清胃促生长素、胃泌素、瘦素、神经肽Y水平。结果:32周龄模型组、64周龄模型组分别与同周龄对照组比较,在胃肠动力学指标差异有统计学意义(P<0.05或P<0.01),血清胃泌素、瘦素、神经肽Y水平差异也均有统计学意义(P<0.05或P<0.01),免疫组化检测结果显示胃促生长素表达有增龄性递减趋势。结论:脾阳虚模型大鼠“运”“化”功能随增龄减弱,与胃促生长素调节胃肠运动和调控胃泌素、瘦素、神经肽Y水平通路机制有关。
Objective: To investigate the age-related changes of “Yun” “Hua ” function in rats with spleen yang deficiency syndrome. Methods: Forty newborn Wistar rats weighing 5.6 ± 0.5 g were randomly divided into 8-week-old model group, 16-week-old model group, 32-week-old model group, 64-week-old model group 4, with 10 in each group. Another 40 rats were used to establish the corresponding control group with the same age 4 groups, 10 in each group. After corresponding weeks of age, the indexes of gastrointestinal motility were detected, the expression of ghrelin was detected by immunohistochemistry, the levels of ghrelin, gastrin, leptin and neuropeptide were detected by enzyme-linked immunosorbent assay and radioimmunoassay Y level. Results: The 32-week-old model group and 64-week-old model group had significant differences in gastrointestinal motility (P <0.05 or P <0.01), serum gastrin, leptin , Neuropeptide Y levels were also statistically significant differences (P <0.05 or P <0.01), immunohistochemical results showed that ghrelin expression increased age-decreasing trend. Conclusion: The function of “Yun” "Hua ’in rats with spleen yang deficiency is weakened with age, which is related to the regulation of gastrointestinal motility and regulation of gastrin, leptin and neuropeptide Y levels by gastric somatotropin.
