颜氏降脂方降脂作用机理实验研究

目的:研究颜氏降脂方的降脂作用机理。方法:将31只大鼠随机分为5组:空白对照组、模型组、颜氏降脂方低剂量组、颜氏降脂方高剂量组、西药组。空白对照组以普通饲料饲喂大鼠,其余组均以高脂饲料饲喂大鼠5周,制作高脂血症大鼠模型。造模成功后,对各组施予不同处理:空白对照组继续以普通饲料饲喂大鼠,余组仍以高脂饲料饲喂,且空白对照组、模型组均以生理盐水灌胃,颜氏降脂方低剂量组、颜氏降脂方高剂量组、西药组分别以中药低剂量药液、中药高剂量药液、辛伐他汀药液灌胃给药。于实验第8周,收集大鼠粪便,测定其中胆汁酸含量。于实验第12周处死大鼠,经腹主动脉采血,测定血清胆固醇(CHO)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)水平。结果:与空白对照组比较,模型组大鼠粪胆汁酸浓度明显升高(P<0.05)。颜氏降脂方高剂量组大鼠粪胆汁酸浓度较模型组明显升高(P<0.05),西药组大鼠粪胆汁酸浓度较模型组明显降低(P<0.05)。与空白对照组比较,模型组大鼠血清CHO、LDL水平明显上升(P<0.05)。与模型组比较,颜氏降脂方高剂量组大鼠血清CHO、LDL水平明显下降(P<0.05);西药组大鼠血清CHO明显下降(P<0.05)。与空白对照组比较,模型组大鼠血清HDL水平明显升高(P<0.05);与模型组比较,西药组大鼠血清HDL水平明显下降(P<0.05)。与空白对照组比较,模型组大鼠血清HDL/CHO比值明显下降(P<0.05);与模型组比较,颜氏降脂方高剂量组大鼠血清HDL/CHO比值较模型组明显上升(P<0.05)。结论:颜氏降脂方可通过增加粪便中胆汁酸的排泄来改善血清脂质含量。 Objective: To study the mechanism of lipid-lowering effect of Yansheng Jiangzhi prescription. Methods: Thirty-one rats were randomly divided into five groups: control group, model group, low dose of Yansheng Jiangzhi prescription group, high dose of Yanshi Jiangzhi prescription group and western medicine group. The blank control group was fed with normal feed, the rest were fed with high-fat diet for 5 weeks to make hyperlipidemic rat model. After successful modeling, different treatments were given to each group: the blank control group continued to feed rats with normal feed, while the remaining groups were still fed with high-fat diet, and the blank control group and model group were given normal saline, Siphonoid low-dose group, Yan’s lipid-lowering high-dose group, western medicine group were low-dose traditional Chinese medicine liquid, Chinese medicine high-dose liquid, simvastatin liquid intragastric administration. At the 8th week of experiment, rat feces were collected and the contents of bile acids were determined. Rats were sacrificed on the 12th week of the experiment, blood was taken from the abdominal aorta, and serum cholesterol (CHO), low density lipoprotein (LDL) and high density lipoprotein (HDL) levels were measured. Results: Compared with the blank control group, the concentration of fecal bile acid in the model group was significantly increased (P <0.05). Compared with the model group, the concentration of fecal bile acids in the high-dose Yanshi Jiangzhi Fang high-dose group was significantly higher than that in the model group (P <0.05). The fecal bile acid concentration in the western medicine group was significantly lower than that in the model group (P <0.05). Compared with the blank control group, serum CHO and LDL levels in the model group increased significantly (P <0.05). Compared with the model group, serum CHO and LDL levels were significantly decreased in the Yanshi Jiangzhi high dose group (P <0.05); serum CHO in the western medicine group was significantly decreased (P <0.05). Compared with the blank control group, serum HDL level in the model group was significantly increased (P <0.05). Compared with the model group, serum HDL level in the western medicine group was significantly decreased (P <0.05). Compared with the blank control group, the serum HDL / CHO ratio in the model group decreased significantly (P <0.05). Compared with the model group, the serum HDL / CHO ratio in the high-dose Yanshi Jiangzhifang group increased significantly compared with the model group (P <0.05). Conclusion: Yan lipid-lowering side can improve serum lipid content by increasing bile acid excretion in feces.
