目的观察维拉帕米(verapanil)对冻僵大鼠血液有形成分的影响。方法实验动物为健康雄性wistar大鼠,随机分为5组,为对照组、冻僵组、干预1组、干预2组和干预3组。对照组于室温20℃下饲养,干预1、2、3组冷暴露前30min分别腹腔注射维拉帕米5、10和20mg/kg,冻僵组和干预组大鼠-30℃环境进行冷暴露80min后复温4h。观察血液WBC、RBC、PLT数量和肛温的变化。结果冻僵组和干预1组WBC数量明显高于对照组(P<0.01),干预2组WBC数量高于对照组(P<0.05),干预3组WBC数量与对照组比较,差异无统计学意义(P>0.05);各组红细胞数量差异无统计学意义(P>0.05);冻僵组PLT数量较对照组显著增高(P<0.01),而干预组PLT数量与对照组比较,差异无统计学意义(P>0.05);冻僵组和干预组肛温均低于31℃,与对照组比较,温差差异P<0.05,干预3组温差明显低于冻僵组(P>0.05)。结论10mg/kg维拉帕米能有效调节重度冻僵大鼠的血液有形成分数量,改善血液循环,防止体温过低的副效应。
Objective To observe the effect of verapanil on the blood components of frozen rats. Methods The experimental animals were healthy male wistar rats, which were randomly divided into 5 groups: control group, freezing group, intervention group 1, intervention group 2 and intervention group 3. The control group was housed at room temperature of 20 ℃. The intervention groups were exposed to verapamil 5, 10 and 20 mg / kg 30 min prior to cold exposure. The cold exposure group and the intervention group were subjected to cold exposure at -30 ℃ After 80min rewarming 4h. Blood WBC, RBC, PLT number and rectal temperature changes. Results The number of WBCs in the frozen group and intervention group 1 was significantly higher than that in the control group (P <0.01), and the WBC count in the intervention group 2 was higher than that in the control group (P <0.05). There was no significant difference in WBC count between the intervention group and the control group (P> 0.05). There was no significant difference in the number of erythrocytes in each group (P> 0.05). The number of PLT in the frozen group was significantly higher than that in the control group (P <0.01), while the number of PLT in the intervention group was significantly lower than that in the control group (P> 0.05). The rectal temperature in the frozen group and the intervention group were both lower than 31 ℃. Compared with the control group, the difference in temperature difference was P <0.05. The difference in temperature between the intervention group and the intervention group was significantly lower than that in the frozen group (P> 0.05). Conclusion 10mg / kg verapamil can effectively regulate the amount of blood components in severely frozen rats, improve blood circulation and prevent hypothermia.
