人精子穿透去透明带地鼠卵试验中受精卵的制备一般采用压卵法,但该技术难度较大,不易掌握,丢失卵子的机会较多,而且在应用上受到局限。我们对这一过程进行了改进,采用甲醇、冰醋酸、水(5∶1∶2)混合固定液,固定、低渗同步法制备受精卵。经过多次试验表明,本方法稳定,重复性好。改进的方法不仅可用于男性学,生殖生物学等临床和研究领域,还可与分子生物学相结合、在分子水平上研究种间遗传物质的传递,减数分裂后基因表达的研究,非整倍体形成机制以及研究外源性因子如某些病毒和癌基因通过生殖细胞进入人类的机制等。因此,它在医学生物学,单倍体遗传学,以及人类遗传学等领域中具有不可估量的应用前景。
Human sperm penetration to the zona pellucida test in the preparation of fertilized eggs are generally used oocyte compression method, but the technology is more difficult to grasp, there is more chance of losing eggs, but also limited in the application. We improved this process by using fixed, hypotonic synchronous method to prepare fertilized eggs using mixed solution of methanol, acetic acid and water (5: 1: 2). After several tests show that the method is stable and reproducible. The improved method can not only be used in clinical and research fields such as male and reproductive biology, but also in combination with molecular biology to study the transmission of interspecific genetic material at molecular level, gene expression after meiosis, The mechanism of ploidy formation and the mechanism of studying exogenous factors such as the entry of certain viruses and oncogenes into human via germ cells. Therefore, it has immeasurable application prospects in the fields of medical biology, haploid genetics, and human genetics.
