目的 探讨阿霉素耐药乳腺癌细胞系MCF 7/ADM的ABC家族基因的表达及其与敏感细胞系MCF 7/S的差异 ,观察中药雄黄对ABC膜转运蛋白超家族中各基因 (mdr 1、mrp、bcrp)表达的影响。 方法 应用RT PCR检测ABC家族基因的转录表达 ,MTT法检测耐药性改变。 结果 ABC家族基因在MCF 7/ADM细胞中均过表达 ,而在MCF 7/S细胞中均未见表达 ;雄黄可下调ABC家族基因的mRNA水平 ,降低化疗药物阿霉素 (ADM)对MCF 7/ADM细胞的IC50 。 结论 乳腺癌MCF 7/ADM细胞获得ADM抗药性 ,与ABC家族基因过表达有密切关系 ;雄黄可部分逆转MCF 7/ADM细胞对ADM的抗药性。
Objective To investigate the expression of ABC family genes in adriamycin-resistant breast cancer cell line MCF 7 / ADM and its difference with the susceptible cell line MCF 7 / S, and to observe the effect of realgar on the expression of various genes in ABC membrane transporter superfamily , Mrp, bcrp) expression. Methods RT-PCR was used to detect the transcriptional expression of ABC family genes. The drug resistance was tested by MTT assay. Results ABC family genes were overexpressed in MCF 7 / ADM cells but not in MCF 7 / S cells. Realgar could down-regulate the mRNA level of ABC family genes and decrease the inhibitory effect of chemotherapeutic drug adriamycin on MCF 7 / / ADM cells. Conclusion ADM resistance was found in breast cancer MCF 7 / ADM cells, which was closely related to the overexpression of ABC family genes. Realgar could partially reverse the drug resistance of MCF 7 / ADM cells to ADM.
