目的探讨激活Notch信号通路对人牙髓细胞增殖与分化的作用。方法选取完整拔除的健康前磨牙与第三磨牙,应用酶消化法进行原代培养与传代培养。观察培养结果。结果第4代B组人牙髓细胞中的Notch信号通路的下游基因Hesl表达量低于C组(P<0.05);第8代,B组人牙髓细胞中的Hesl表达量与C组比较,差异有统计学意义(P<0.05);第10代,B组人牙髓细胞中的Hesl表达量与C组比较,差异有统计学意义(P<0.05)。结论 Notch信号通路是进化过程中的信号转导机制,能够细胞间作用对其命运进行控制,牙髓细胞存在较高的增殖以及多向分化的成体干细胞,在组织中起到非常重要的作用。
Objective To investigate the effect of activating Notch signaling pathway on the proliferation and differentiation of human dental pulp cells. Methods The healthy premolar and the third molars with complete removal were selected and primary culture and subculture were performed by enzyme digestion. Observe the culture results. Results The expression level of Hesl in Notch signaling pathway in human dental pulp cells of the 4th generation was lower than that of the C group (P <0.05). Compared with the C group, the expression of Hesl in the 8th and the B group of human dental pulp cells , The difference was statistically significant (P <0.05). The expression level of Hesl in the human dental pulp cells of the 10th generation and the B group was significantly different from that of the C group (P <0.05). Conclusion Notch signaling pathway is a signal transduction mechanism during evolution. It can control the fate of the Notch signaling pathway. The proliferation of pulp cells and the differentiation of adult stem cells play a very important role in the tissue.
