目的研究干扰素-α联合姜黄素对B-NHL淋巴瘤Raji细胞的诱导凋亡作用及其作用机制。方法以不同浓度干扰素-α(500、1000、2000/L),不同浓度的姜黄素6.25、12.5、25μmol/L)和IC502000U/L的干扰素-α联合不同浓度姜黄素作用于体外培养的Raji细胞,观察细胞生长状态变化。MTT法检测细胞生长的抑制率,应用流式细胞仪法观察细胞凋亡,Western blot法检测24h,姜黄素IC5025μmol/L联合2000U/L的干扰素-α作用于caspase6、caspase8、caspase9的蛋白表达。结果干扰素-α与姜黄素联合应用可显著抑制Raji细胞的生长.诱导细胞发生凋亡,两者具有协同效应。凋亡蛋白caspase6、 caspase8、caspase9表达显著增高,其作用呈现出明显的量-效与时-效关系。结论干扰素-α与姜黄素联合应用对B-NHL淋巴瘤Raji细胞具有明显的增生抑制作用,促进凋亡蛋白的表达及诱导细胞凋亡,可能是其重要作用机制之一。
Objective To study the effect of interferon-α combined with curcumin on the apoptosis of B-NHL lymphoma Raji cells and its mechanism. Methods Different concentrations of interferon-α (500,1000,2000 / L), different concentrations of curcumin 6.25,12.5,25μmol / L) and IC502000U / L interferon-α combined with different concentrations of curcumin Raji cells were cultured in vitro to observe the changes of cell growth status. The inhibitory rate of cell growth was detected by MTT assay. The apoptosis was observed by flow cytometry. The protein expression of caspase6, caspase8 and caspase9 was detected by Western blot after 24h, curcumin IC5025μmol / L combined with 2000U / L interferon-α . Results Interferon-α combined with curcumin significantly inhibited the growth of Raji cells. Induction of cell apoptosis, the two have a synergistic effect. The expressions of caspase6, caspase8 and caspase9 were significantly increased, and their effects were dose-response and time-effect. Conclusions The combination of interferon-α and curcumin can inhibit the proliferation of B-NHL lymphoma Raji cells, promote the expression of apoptosis protein and induce the apoptosis of Raji cells, which may be one of the important mechanisms.
