目的:通过对不同盐碱胁迫下红芪种子萌发及幼苗生理特性的研究,寻找提高红芪种子及幼苗在不同盐碱胁迫下抗性能力的途径。方法:测定不同盐碱胁迫下红芪种子的发芽势(Gv)、发芽率(Gr)、发芽指数(Gi)和相对盐害率,并对红芪幼苗的叶绿素含量、可溶性蛋白质含量、质膜透性、超氧化物歧化酶(SOD)和过氧化物酶(POD)的活性进行了测定。结果:3种盐碱胁迫处理均引起红芪种子发芽率下降,发芽势、发芽指数降低,同时相对盐害率升高,而且随着盐碱胁迫浓度的增加,抑制作用增强。但对种子的胁迫影响大小为Na2CO3﹥Na2SO4﹥NaCl。随着3种盐碱处理浓度的增大,红芪幼苗中叶绿素含量逐渐较少,可溶性蛋白质的含量也逐渐降低,二者都与盐碱处理浓度呈负相关性,但质膜透性逐渐增大与盐碱处理浓度呈正相关性,SOD,POD活性均不同程度的表现为先上升后下降的趋势,均在Na2SO4和Na2CO3浓度为25 mmol·L-1时出现最大值,而NaCl在浓度为50 mmol·L-1时出现最大值。结论:3种盐碱对红芪种子萌发和幼苗生理指标的抑制效应为Na2CO3﹥Na2SO4﹥NaCl。
OBJECTIVE: To find ways to improve the resistance of Radix Hedysari seeds and seedlings under different saline-alkali stress by studying the seed germination and seedling physiological characteristics under different salt-alkali stress. Methods: The germination potential (Gv), germination rate (Gr), germination index (Gi) and relative salt damage rate of Radix Astragali were determined under different salinity-alkalinity stress. Chlorophyll content, soluble protein content, Permeability, superoxide dismutase (SOD) and peroxidase (POD) activity were measured. Results: The three kinds of salt-alkali stress all caused the germination rate of Red Hedysarum seeds decreased, the germination potential and germination index decreased, while the relative salt damage rate increased, and with the increase of saline-alkali concentration, the inhibition increased. However, the effect of stress on seed size is Na2CO3> Na2SO4> NaCl. With the increase of the concentration of three kinds of saline-alkali, the content of chlorophyll gradually decreased and the content of soluble protein decreased gradually. Both of them were negatively correlated with the concentration of saline-alkali, but the permeability of the plasma membrane gradually increased The results showed that there was a positive correlation between the concentration of salinity and alkali and the concentration of salinity-alkalinity treatment. The activities of SOD and POD increased at first and then decreased, both of which were highest when the concentration of Na2SO4 and Na2CO3 was 25 mmol·L-1, 50 mmol · L-1 when the maximum value. CONCLUSION: The inhibitory effects of three kinds of saline-alkali on seed germination and seedling physiological characteristics of Radix Astragali were Na2CO3> Na2SO4> NaCl.
