目的:考察蛇葡萄素钠单用及与卡铂合用对人肺腺癌GLC-82细胞增值的抑制作用,初步探讨蛇葡萄素钠抑瘤作用机制。方法:应用MTT法考察蛇葡萄素钠单用及与卡铂合用对人肺腺癌GLC-82细胞的细胞毒作用;使用流式细胞仪检测凋亡相关基因Caspase-3的表达。结果:MTT实验结果表明,蛇葡萄素钠在浓度为12.5~200μg/ml的剂量范围内对GLC-82细胞的增殖有浓度依赖性的抑制作用;卡铂在浓度为3.13~100μg/ml的剂量范围内对GLC-82细胞的增值有浓度依赖性的抑制作用;25、50μg/ml的蛇葡萄素钠对卡铂25μg/ml抑制GLC-82细胞增殖的作用具有协同效应;流式细胞仪检测结果表明,12.5~50μg/ml的蛇葡萄素钠与卡铂25μg/ml联合用药组作用于GLC-82细胞12 h后,Caspase-3表达上调,且有浓度依赖性。结论:蛇葡萄素钠单用对GLC-82细胞的增殖具有一定的抑制作用,与卡铂合用,对GLC-82细胞增殖具有协同抑制效应;蛇葡萄素钠诱导细胞凋亡的机制之一可能是通过激活细胞内的Caspase-3,从而促进凋亡的发生。
OBJECTIVE: To investigate the inhibitory effect of combined use of snakecein sodium and carboplatin on the proliferation of human lung adenocarcinoma GLC-82 cells and to explore the mechanism of the inhibitory effect of snakecein sodium. CONCLUSION: The combination of carbachol and venlafaxine could inhibit the proliferation of GLC-82 cells in vitro. One of the mechanisms of apoptosis induced by sodium phytic acid may be one of the mechanisms Is through the activation of intracellular Caspase-3, thereby promoting the occurrence of apoptosis.
