目的:研究c-Met在胃癌组织中的表达及其临床意义。方法:收集2011年1月至2011年12月盐城市第一、三人民医院外科接诊、手术切除并经病理证实的胃癌石蜡标本69例,另取同期胃黏膜不典型增生组织标本20例以及正常胃黏膜标本20例作对照。采用免疫组织化学法检测胃癌组织中c-Met蛋白的表达,采用real-time PCR法检测15例手术切除的胃癌和癌旁组织中c-Met mRNA的表达。采用Spearman秩相关分析评估c-Met蛋白与胃癌标志物骨桥蛋白(osteopontin,OPN)、基质金属蛋白酶(matrix metalloproteinase,MMP)表达的关系。结果:c-Met蛋白在胃癌组织中的阳性表达率为65.2%,明显高于胃黏膜不典型增生组织(30%)及正常胃黏膜组织(20%,均P<0.01);c-Met蛋白表达与胃癌的淋巴结转移、临床TNM分期相关(P<0.01)。胃癌组织中c-Met mRNA的表达显著高于正常胃黏膜组织[(0.20±0.12)vs(0.03±0.02),P<0.01]。OPN、MMP-9蛋白的表达与胃癌的浸润、临床TNM分期、淋巴结转移相关,且c-Met蛋白的表达与OPN、MMP-9的表达呈正相关(P<0.05)。结论:c-Met在胃癌组织中高表达,在胃癌的发生、发展过程中具有重要的作用,与OPN、MMP-9可能存在一定的协同性。
Objective: To study the expression of c-Met in gastric carcinoma and its clinical significance. Methods: From January 2011 to December 2011, 69 cases of gastric cancer paraffin specimens surgically and surgically removed from the first and third People’s Hospital of Yancheng from January 2011 to December 2011 were collected. Twenty specimens of gastric dysplasia tissues were collected from January 2011 to December 2011. 20 cases of normal gastric mucosa as a control. The expression of c-Met protein in gastric cancer tissues was detected by immunohistochemistry. The expression of c-Met mRNA in 15 cases of resected gastric cancer tissues and adjacent normal tissues was detected by real-time PCR. Spearman rank correlation analysis was used to evaluate the relationship between c-Met protein and the expression of osteopontin (OPN) and matrix metalloproteinase (MMP). Results: The positive rate of c-Met protein in gastric cancer was 65.2%, which was significantly higher than that in gastric atypical hyperplasia (30%) and normal gastric mucosa (20%, both P <0.01) The expression was correlated with lymph node metastasis and TNM staging of gastric cancer (P <0.01). The expression of c-Met mRNA in gastric cancer tissues was significantly higher than that in normal gastric mucosa [(0.20 ± 0.12) vs (0.03 ± 0.02), P <0.01]. The expression of OPN and MMP-9 was correlated with the infiltration of gastric cancer, TNM staging and lymph node metastasis. The expression of c-Met protein was positively correlated with the expression of OPN and MMP-9 (P <0.05). Conclusion: The high expression of c-Met in gastric cancer tissue plays an important role in the development and progression of gastric cancer. There may be some synergy with OPN and MMP-9.
