目的:研究麻黄及其与桂枝配伍前后麻黄生物碱在大鼠体内组织分布变化的特征,并对二者的分布情况进行对比研究。方法:以金刚烷胺为内标,采用UPLC-MS/MS法测定大鼠灌胃给予麻黄单煎液及麻黄-桂枝药对水煎液后三个时间点(0.25、1.5、3h)组织样品中5种麻黄生物碱的浓度。结果:与麻黄组相比,给药后0.25h,不同程度的降低去甲基麻黄碱、麻黄碱和伪麻黄碱在各组织的含量,增加了去甲基伪麻黄碱和甲基麻黄碱在各组织的含量;在分布的平衡期(1.5h),麻黄桂枝组均能显著提高麻黄类生物碱在肺组织中的含量,提高去甲基麻黄碱、去甲基伪麻黄碱和甲基麻黄碱在肾组织中的含量;在给药后3h,麻黄桂枝组均能显著提高麻黄类生物碱在脑组织中的含量,显著提高去甲基麻黄碱、麻黄碱和伪麻黄碱在肾组织中的含量,显著降低去甲基麻黄碱、麻黄碱、伪麻黄碱和甲基麻黄碱在心组织的含量。结论:桂枝增加了麻黄类生物碱在大鼠脑组织中的分布,增加去甲基麻黄碱、麻黄碱和伪麻黄碱在肾中的分布;降低了去甲基麻黄碱、麻黄碱、伪麻黄碱和甲基麻黄碱在心组织的分布。
OBJECTIVE: To study the changes of tissue distribution of ephedra alkaloids before and after the compatibility of ephedra and Guizhi, and to compare their distribution. Methods: Amantadine was used as internal standard. Results: Compared with the ephedra group, the content of demethoxymethane, ephedrine and pseudoephedrine were decreased in all tissues at 0.25h after administration, which increased the content of demethoxy pseudoephedrine and methylephedrine in different tissues ; In the distribution of the balance period (1.5h), Ephedra Guizhi group can significantly improve the Ephedra alkaloids in the lung tissue content, to improve the demethoxymethane, demethylated pseudoephedrine and methyephedrine in the kidney tissue ; 3 h after administration, the group of Ephedra Guizhi can significantly improve the content of ephedrine alkaloids in the brain tissue, significantly increased the content of demethoxymethane, ephedrine and pseudoephedrine in the kidney tissue, significantly reduced to Methylephedrine, ephedrine, pseudoephedrine and methylephedrine in cardiac tissue. Conclusion: Guizhi increased the distribution of ephedrine alkaloids in rat brain tissue, increased the distribution of demethoxy ephedrine, ephedrine and pseudoephedrine in the kidneys, decreased the contents of demethylethylene, ephedrine, pseudoephedrine and Ephedrine distribution in heart tissue.
