目的:建立HPLC法测定齐墩果酸缓释微丸胶囊中齐墩果酸含量的方法。方法:以Kromasil C_(18)(250 mm×4.6 mm,5μm)为色谱柱,流动相为甲醇-0.3%三乙胺溶液(85:15),检测波长为210 nm,流速为1.0 ml·min~(-1),柱温为30℃,进样量为20μl。结果:齐墩果酸在50.12~501.20μg·ml~(-1)范围内线性关系良好(r=0.999 6),平均回收率为99.72%(RSD=0.9%,n=6)。结论:本方法简便、准确、专属性强,可用于该制剂的含量测定。
Objective: To establish a HPLC method for the determination of oleanolic acid in sustained release pellets. Methods: Kromasil C 18 (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of methanol-0.3% triethylamine (85:15), the detection wavelength was 210 nm and the flow rate was 1.0 ml · min ~ (-1). The column temperature was 30 ℃ and the injection volume was 20μl. Results: Oleanolic acid showed good linearity (r = 0.999 6) in the range of 50.12-501.20 μg · ml -1 with an average recovery of 99.72% (RSD = 0.9%, n = 6). Conclusion: The method is simple, accurate, specific and can be used for the determination of the content of the preparation.
