目的通过观察酪氨酸磷酸化的胰岛素受体底物(P-Ikbα)在胰岛素抵抗小鼠肝脏、胰腺的表达随时间变化的差异性,了解细胞水平胰岛素抵抗与系统胰岛素抵抗的关系。方法 C57小鼠随机分为高脂组、胰岛素兴奋组和对照组,均尾静脉监测血糖、体重及胰岛素水平变化。取肝脏、胰腺应用Western Blot方法检测P-Ikbα的表达。结果与对照组相比,高脂组和胰岛素兴奋组肝脏、胰腺P-Ikbα蛋白表达随时间延长而减少。血清胰岛素水平在造模第7天升高,而肝脏的IRS-1酪氨酸磷酸化在第14天末低于正常对照组,胰腺在第21天末低于正常组。结论肝脏IRS-1酪氨酸磷酸化水平的降低早于胰腺;血清胰岛素升高发生时间早于细胞水平胰岛素抵抗发生时间。
OBJECTIVE: To investigate the relationship between insulin resistance and insulin resistance at the cellular level by observing the difference of the expression of tyrosine phosphorylated P-Ikbα in liver and pancreas of mice with insulin resistance over time. Methods C57 mice were randomly divided into hyperlipidemia group, insulin excitement group and control group. Blood glucose, body weight and insulin levels were monitored by tail vein. The liver and pancreas were used to detect the expression of P-Ikbα by Western Blot. Results Compared with the control group, the expression of P-Ikbα protein in the liver and pancreas decreased in the hyperlipidemic and insulin-stimulated groups with time prolonging. Serum insulin levels increased on the 7th day of modeling, while IRS-1 tyrosine phosphorylation in the liver was lower than the normal control group on the 14th day. The pancreas was lower than the normal group on the 21st day. Conclusions The decrease of IRS-1 tyrosine phosphorylation in liver is earlier than that in pancreas. The serum insulin level rises earlier than the time of cell level insulin resistance.
