目的 研究人类疱疹病毒 6、7型 (humanherpesvirus 6and 7,HHV 6、HHV 7)体外感染对免疫功能的影响。方法 采用生物活性法或ELISA法分别检测了HHV 7Glasgow、YY5 2株病毒及HHV 6GS株感染脐血单个核细胞 (CBMCs) ,培养上清中细胞因子TNF α、IFN γ、IL 10、IL 12P70水平的动态变化。结果 3株病毒感染CBMCs后前 2天均轻度抑制IFN γ产生 ,然后此抑制作用逐渐消失 ;3株病毒均能平稳诱导IL 12P70产生 ,但GS组诱生水平低于Glasgow及YY5组 ;3株病毒在接种后早期都能诱生IL 10 ,GS诱生IL 10水平高于Glasgow、YY5株 ;3株病毒均能较强地诱导CBMCs产生TNF α ,但到达峰值时间Glasgow、YY5迟于GS株。结论 HHV 6、HHV 7感染早期都可以通过影响细胞因子的分泌影响免疫功能 ,但HHV 6影响程度强于HHV 7。
Objective To study the effects of human herpesvirus 6 and 7 (HHV 6 and HHV 7) on the immune function in vitro. Methods Cord blood mononuclear cells (CBMCs) were infected with HGV 7Glasgow, YY5 2 and HHV 6GS strains by bioactivity or ELISA. The levels of cytokines TNFα, IFNγ, IL 10 and IL 12P70 Dynamic changes. Results The IFNγ production was mildly inhibited in the first two days after the three strains of CBMCs were infected, and then the inhibition gradually disappeared. All of the three viruses could induce the IL 12P70 production steadily, but the induced levels in GS group were lower than those in Glasgow and YY5 groups. The IL-10 was induced in the early stage of inoculation, and the level of IL-10 induced by GS was higher than that in Glasgow and YY5 strains. All of the three viruses could induce the production of TNFα in CBMCs. However, the peak time of Glasgow and YY5 was delayed than that of GS Strain. Conclusions Both HHV 6 and HHV 7 could affect the immune function by affecting the secretion of cytokines in the early stage of infection, but the effect of HHV 6 is stronger than that of HHV 7.
