目的分析余姚市手足口病的病原谱构成,了解EV71的基因特征。方法收集1 827份手足口病病例标本,用实时荧光PCR检测并分型,样品进行肠道病毒71型(EV71)VP1基因的扩增和核苷酸序列测定,并用DNAstar和Blast软件对测序结果进行比对分析。结果 2010年-2013年1 827份标本中EV71和Cox A16的检出率分别为37.71%、24.25%,其他肠道病毒的检出率为22.28%;EV71 VP1蛋白氨基酸序列相似性为98.65%~100.00%,11株EV71基因分型属于C4a基因亚型。结论 2010年-2012年余姚市手足口病流行的主要病原体为EV71和Cox A16,2013年以其他肠道病毒为主。流行的EV71属C4a基因亚型,并具有高度相似性。
Objective To analyze the pathogenic spectrum of hand-foot-mouth disease in Yuyao, and to understand the genetic characteristics of EV71. Methods A total of 1 827 samples of hand, foot and mouth disease were collected and detected by real-time fluorescence PCR. The samples were used to amplify and sequence the VP1 gene of enterovirus 71 (EV71). DNAstar and Blast software were used to analyze the sequencing results For comparison analysis. Results The positive rates of EV71 and Cox A16 were 37.71% and 24.25% respectively among the 1 827 specimens from 2010 to 2013. The positive rate of other enterovirus was 22.28%. The amino acid sequence of EV71 VP1 protein was 98.65% 100.00%, 11 EV71 genotypes belong to C4a gene subtype. Conclusion From 2010 to 2012, the major pathogens of HFMD in Yuyao are EV71 and Cox A16. The popular EV71 is a subtype of C4a and has a high degree of similarity.
