马铃薯是世界上重要的栽培作物,俄罗斯品种因其类型丰富而闻名,为了将俄引资源中的优良基因快速用于我国马铃薯品种改良,本研究利用新近发表的SSR分子标记,对52份从俄罗斯引进的马铃薯品种进行群体结构性研究并构建其分子身份证。结果显示,从100份引物中筛选出的23对的目标条带清楚、多态性高且扩增稳定的引物,在俄罗斯马铃薯群体中共检测到86个多态性目标条带,每对引物可观测等位基因数为2~5个,平均等位基因数为3.7个。引物位点的多态信息含量指数(PIC)变幅为0.033 9~0.688 4,平均多态性信息含量为0.528 5。品种间特异性指数差异较大,介于150.762~273.970之间,平均为184.921。俄罗斯马铃薯材料蕴含了比较丰富的遗传变异,显示了较高水平的基因多样性。在多样性研究基础上,利用STL029、STPOAC58、STM1106、STL047、STM0051、STM1030及STL004共7对引物构建了63份种质资源的分子身份证,可有效区分各品种。上述结果为利用俄引资源改良黑龙江省马铃薯品种提供了分子遗传学信息
Potato is one of the most important cultivated crops in the world. Russian varieties are well-known for their rich variety. In order to rapidly apply the excellent genes from Russian sources to the improvement of potato varieties in our country, the present study utilized the newly published SSR markers, The introduction of potato varieties for population structural studies and the construction of its molecular ID card. The results showed that, of the 23 pairs of primers selected from 100 primers, the target bands were clear, with high polymorphism and stable amplification. A total of 86 polymorphic bands were detected in the Russian potato population, and each pair of primers The number of observed alleles was 2 to 5, and the average number of alleles was 3.7. The polymorphism information content index (PIC) of primer loci ranged from 0.033 9 to 0.688 4, and the average polymorphism information content was 0.528 5. Differences in the specific index between cultivars ranged from 150.762 to 273.970 with an average of 184.921. Russian potato material contains a wealth of genetic variation, showing a high level of genetic diversity. Based on the diversity studies, 63 germplasm molecular ID cards were constructed using 7 pairs of primers of STL029, STPOAC58, STM1106, STL047, STM0051, STM1030 and STL004, which can effectively distinguish the varieties.
